Jayne began to move into more mainstream programming with three series of the fashion show She's Gotta Have It. She went on to present some of the BBC's Holiday programmes, to destinations across the globe before co-hosting the LA Pool Party series with Lisa Snowdon, which won a TRIC Award.

In 2005, Jayne presented the dating show What Women Want, before co-presenting the seventh series of Robot Wars, alongside Craig Charles and The Games with Jamie Theakston. Additionally, she appeared on Celebrity Love Island and became emotionally involved with footballer Lee Sharpe. The pair did not become a couple, however, despite the producers of the show sending them on a trip in a 'make or break' boat. She eventually went on to win the show, alongside Fran Cosgrave. In 2006 she returned to Fiji to host Celebrity Love Island's sister show Aftersun on ITV2.

Other credits include coverage of Live 8 for commercial radio, ITV2's coverage of the British Comedy Awards and Soapstar Superstar, reporting for GMTV and Richard and Judy and appearances as a 'talking head' in a number of celebrity-based documentaries.

A committed follower of yoga for over ten years, in 2005 Jayne released the hit DVD Jayne Middlemiss - Love Yoga, and then worked on a second fitness release. In 2006, Jayne narrated a documentary about a yoga retreat for the BBC, and she also appeared in A beginners guide to...Yoga for Channel 4 (2007).

In 2008 Jayne appeared on Ant and Dec's Saturday Night Takeaway, and the following year took part in This Morning and Something for the Weekend. In 2009 she also won Celebrity MasterChef on BBC One, and in 2010 began her role as presenter on Live from Studio Five.
